Ultrasonic‐Assisted Synthesis of Spiro-Indeno-Thiazines: A Green, Simple and One-pot Approach

Spiro-thiazines, are very attractive target for combinatorial library synthesis due to their structure activity relationship and members of an important class of N and S containing six-membered heterocycles [1]. They are widely used as key building blocks in the field of pharmaceutical agents and drugs. As an extension of these studies [2, 3], a useful and green strategy using ultrasonic waves via a four-component reaction between malononitrile, ninhydrin, tetramethylguanidine and heterocumulene has been reported for the synthesis of spiro(indeneo-thiazine) in water solvent, room temperature and short reaction time (30 min). After optimizing the reaction in H2O solvent, it was investigated under ultrasonic conditions for 30 min using 1.0 mmol of tetramethylguanidine 1, 1.5 mmol of various isothiocyanates 2, 1.5 mmol of ninhydrin 3, and 1.0 mmol of malononitrile 4 (see Table 1). The use of simple and available starting materials, good to high yields, no need for column chromatography and short reaction time are important features of this protocol. The synthesized compounds have been identified using IR, Mass and NMR spectroscopy methods.
