A Five-Component Cascade Synthesis of Highly Functionalized α-Diimine Heterocycles Using Nitro Ketene Aminals

Since their discovery in 1990s, α-diimine complexes of nickel and palladium are one of the most studied late-transition organometallics as single-site catalysts that are used for polymerization of ethylene, α-olefins and certain polar functionalized olefins.
In this work, an effective five-component synthesis of completely novel bis α-diimine structures including fused heterocycles; imidazo[1,2-a]pyridine, pyrido[1,2-a]pyrimidine and pyrido[1,3]diazepine is reported. The starting materials used involve glyoxal 1, cyanoacetohydrazide 2, aromatic aldehydes 3, 1,1-bis(methylthio)-2-nitroethylene 4, and various diamines 5. The reactions were completed within 24 h in EtOH/H2O under acidic conditions at reflux temperature. (Scheme 1 shows the synthesis pathway of desired products).
1H, 13C NMR and IR spectra of the crude products confirmed the synthesis of suggested structures.
As a result of this effort, new heterocyclic compounds were obtained with good yields and it is expected that in the future this work can be done using pyruvic aldehyde, diacetyl and other dicarbonyl compounds instead of glyoxal. The advantage of this approach is characterized by available and affordable starting components, simplicity of operation, use of environmentally safe solvents and high atomic economy.
