An Innovative Aqueous One-Pot Route for Synthesis of Indene-Connected Hybrid Pyrroles

Pyrroles, owing to their broad presence in natural products and pharmaceuticals, constitute highly valuable heterocyclic scaffolds; therefore, developing sustainable methods for their construction is of considerable importance [1-2]. Addressing this need, the present study introduces an efficient and environmentally benign strategy for synthesizing nitrogen-containing indene-pyrrole heterocycles. This one-pot protocol employs ninhydrin, acetophenone, primary amines, and ethyl acetoacetate in water without any catalyst, aligning well with green chemistry principles by eliminating toxic organic solvents [3] (Scheme1). The transformation proceeds through the concurrent formation of multiple C–N and C–C bonds under mild conditions, affording pyrrole derivatives in high yields. Overall, this approach offers a green, practical, and efficient route to indene-pyrrole frameworks with potential relevance in organic and medicinal chemistry.
