Electrochemical oxidation of mesalazine in aqueous and non-aqueous media: Pathways to tetramer formation and olsalazine synthesis

In this study, the electrochemical synthesis of the anti-inflammatory drug olsalazine (OSZ) was investigated via the oxidative dimerization of mesalazine (MSZ). The oxidation of MSZ was carried out in both aqueous and non-aqueous media using cyclic voltammetry and controlled-potential coulometry to determine the optimal conditions for dimer formation. The results revealed that MSZ acts as a nucleophile, reacting with the para-quinonimine species generated from its own oxidation (MSZox), leading to the formation of the corresponding dimer. Two reaction pathways were identified for dimerization and tetramer formation: one involving C–N coupling and the other involving N–N coupling, ultimately resulting in the synthesis of OSZ
