Design of a Smart Biocompatible Chitosan–Hyaluronic Acid–Based Hydrogel for Transdermal Insulin Delivery

Diabetes mellitus requires precise blood glucose control. Insulin is the primary treatment for type 1 and advanced type 2 diabetes [1]. The invasiveness and low patient compliance of subcutaneous injections necessitate the development of non-invasive delivery systems [2]. Smart transdermal drug delivery systems have attracted increasing attention due to their improved patient compatibility and capability for controlled drug release. In the present study, a smart transdermal hydrogel system incorporating glucose oxidase was synthesized via ionic gelation to enable controlled and glucose-mediated insulin release. The hydrogel was formulated using biocompatible polymers, including chitosan, hyaluronic acid, and gelatin. Oxalic acid was employed as a crosslinking agent, while glycerol was incorporated to enhance mechanical properties, resulting in a stable and flexible hydrogel structure. Insulin and glucose oxidase were simultaneously loaded into the hydrogel network. Enzymatic activity in the presence of glucose induced structural changes within the polymeric network, leading to increased permeability. Swelling and structural stability tests confirmed the appropriate performance of the system. Drug release studies conducted using UV–Vis spectroscopy demonstrated that approximately 25% of the loaded insulin was released within 24 hours. Moreover, an increase in glucose concentration led to a higher insulin release rate, indicating the smart and self-regulating behavior of the system. Overall, this glucose-responsive hydrogel, with its controlled insulin release capability, provides a novel platform for the development of non-invasive and smart insulin delivery systems. The findings of this study represent a significant step toward personalized diabetes treatment and demonstrate strong potential for future clinical applications.
