| Hexagonal boron nitride (hBN) nanostructures have attracted increasing interest as advanced nanocarriers for cancer drug delivery due to their outstanding chemical and thermal stability, high surface area, and favorable biocompatibility. However, the intrinsic inertness and limited dispersibility of pristine hBN restrict its biomedical applicability. In this work, hBN was rationally engineered through surface modification with poly(cyclodextrin–citric acid), a biocompatible polymeric network that enhances aqueous stability and provides abundant functional groups for efficient drug encapsulation. To enable active tumor targeting, folic acid was covalently conjugated to the modified hBN nanoplatform, exploiting the overexpression of folate receptors in colon cancer cells. Irinotecan, a first-line chemotherapeutic agent for colorectal cancer, was successfully loaded into the functionalized hBN system via host–guest interactions and surface adsorption. The resulting nanocarrier exhibited high drug loading efficiency and controlled release behavior, contributing to improved therapeutic performance. In vitro evaluation on colon cancer cell models demonstrated enhanced cellular uptake mediated by folate receptor targeting, leading to significantly increased cytotoxicity compared to free irinotecan. Moreover, the nanoplatform showed reduced nonspecific interactions, suggesting a potential decrease in systemic side effects. Overall, the synergistic integration of polymer modification, receptor-mediated targeting, and efficient drug delivery highlights the potential of poly(cyclodextrin–citric acid)-modified hBN as a multifunctional nanoplatform for precision colorectal cancer therapy. This study provides valuable insights into the design of hBN-based nanostructures for targeted anticancer drug delivery and supports their future development toward translational nanomedicine applications [1]. |
